A study of ion-exchange chromatography in an expanded bed for bovine albumin recovery BABT

In the present work, the effect of bed expansion on BSA adsorption on Amberlite IRA 410 ion-exchange resin was studied. The hydrodynamic behavior of an expanded bed adsorption column on effects of the biomolecules and salt addition and temperature were studied to optimize the conditions for BSA recovery on ion-exchange resin. Residence time distribution showed that HEPT, axial dispersion and the Pecletl number increased with temperature and bed height, bed voidage and linear velocity. The binding capacity of the resin increased with bed height. The Amberlite IRA 410 ion-exchange showed an affinity for BSA with a recovery yield of 78.36 % of total protein.

O conhecimento da dinâmica dos nutrientes adicionados no solo via adubação é crucial para o adequado manejo da nutrição das plantas, bem como para avaliar riscos de contaminação do solo e das águas subterrâneas com a aplicação de efluentes. O estudo teve como objetivo avaliar a dinâmica do deslocamento de íons potássicos em amostras de um Latossolo Amarelo muito argiloso, caulinítico, que é a classe de solo dominante na Amazônia Central. Foram determinados os parâmetros de transporte de solutos em amostras de solos de uma camada superficial (0-20 cm) e de uma camada subsuperficial (20-40 cm), por meio de ensaios de retardo (breakthrough curves - BTC). When doing researches on solute dynamics in porous medium, the knowledge of medium characteristics and percolating liquids, as well as of external factors is very important. An important external factor is temperature and, in this sense, our purpose was determining potassium and nitrate transport parameters for different values of temperature, in miscible displacement experiments. Evaluated parameters were retardation factor (R), diffusion/dispersion coefficient (D) and dispersivity, at ambient temperature (25 up to 28 ºC), 40 ºC and 50 ºC. ABSTRACT This study aimed to determine some transport parameters of potassium in soil columns by miscible displacement of effluent from a Sewage Treatment Plant (STP). This effluent was collected at different stages of treatment, and potassium chloride solutions were prepared with the same K pulse concentration as the effluent. They were used to conduct a miscible displacement experiment of potassium in soil columns of dystrophic Red Latosol. Both types of solutions contained concentrations of 30 and 32 mg L-1 of potassium.
